Matching methods for observational microarray studies

چکیده
MOTIVATION We address the problem of identifying differentially expressed genes between two conditions in the scenario where the data arise from an observational study, in which confounding factors are likely to be present. RESULTS We suggest to use matching methods to balance two groups of observed cases on measured covariates, and to identify differentially expressed genes using a test suited to matched data. We illustrate this approach on two microarray studies: the first study consists of data from patients with two cancer subtypes, and the second study consists of data from AMKL patients with and without Down syndrome. AVAILABILITY R code (www.r-project.org) for implementing our approach is included as Supplementary Material. SUPPLEMENTARY INFORMATION Supplementary data are available at Bioinformatics online.

Background and Objective: Studies in the health sciences is comprised of observational and intervention. A major part of health sciences research has been allocated to the observational studies. Designing and doing studies based on scientific guidelines that include the entire process, leads to studies validation and also results can be generalized to the community. Thus, for standardizing ...
متن کاملAn Introduction to Instrumental Variables
Instrumental variables (IVs) are used to control for confounding and measurement error in observational studies. They allow for the possibility of making causal inferences with observational data. Like propensity scores, IVs can adjust for both observed and unobserved confounding effects. Other methods of adjusting for confounding effects, which include stratification, matching and multiple reg...
متن کاملMatching and Propensity Scores
The popularity of matching techniques has increased considerably during the last decades. They are mainly used for matching treatment and control units in order to estimate causal treatment effects from observational studies or for integrating two or more data sets that share a common subset of covariates. In focusing on causal inference with observational studies, we discuss multivariate match...
متن کاملOn Applying String Matching in Searching Unique Oligonucleotides
In this paper we consider using string matching in finding unique, gene specific oligonucleotides, which can be used for example in genomic research involving polymerase chain reaction, gene cloning, microarray technology or antisense DNA studies. Because of the typically huge size of genomes, the practical speed of the method used is important. We tested both exact and approximate string match...
متن کاملMatching methods for causal inference: Designing observational studies
Much research in the social sciences attempts to estimate the effect of some intervention or “treatment” such as a school dropout prevention program or television watching. However, particularly in the social sciences, it is generally not possible to randomly assign units to receive the treatment condition or the control condition, and thus the resulting data are observational, where we simply ...
